Advances in molecular technology have increased the speed and accuracy of PCR testing, however, accurate extraction of DNA and RNA have remained largely unchanged. PCRopsis is a novel, direct to PCR reagent that will give your laboratory the ability to reduce the time, steps, and consumables needed for RNA/DNA extraction without losing the sensitivity of traditional extraction.  Extractions that historically require over 20 steps and 30-90 minutes of extraction processing can in most cases be reduced to 5 steps and 5-15 minutes incubation.

Common Workflows using PCRopsis

UTI Extraction from Urine Sample

  • Prepare working reagent by adding PCRopsis Support to PCRopsis Reagent RVD with RVD Enhancer (reagent mixture)
  • Centrifuge urine sample ~10 minutes in aliquot tube or 96 deep well plate
  • Remove supernatant leaving less than 500ul of urine over the pellet
  • Mix 20uL of remaining sample with 20uL reagent mixture (sample mixture)
  • Heat sample mixture for ~10 minutes at 95°C

Incubation Time: ~10 mins

COVID/RPP-swab in VTM, Oral Rinse, Saliva

  • Thoroughly mix PCRopsis™ Reagent RVD-Enhancer to ensure homogeneity,
  • Mix 1 volume of sample (20 μL) with 1 volume of Reagent RVD-Enhancer (20 μL)
  • Pipette up & down to ensure complete mixing and then seal plate.
  • Heat diluted sample at 95°C for ~10 mins and let cool at room temperature for 10~20 seconds

  • Centrifuge urine sample ~10 minutes in aliquot tube or 96 deep well plate
  • Remove supernatant leaving less than 500ul of urine over the pellet (ideally only 100 uL)
  • Mix 20uL of remaining sample with 20uL reagent mixture or in ratio of 1:1 (sample mixture)
  • Heat sample mixture for ~10-15 minutes at 95°C then let cool for 10-20 seconds

Incubation Time: ~10-15 mins

Fungal/Yeast-Swab in transport media

  • Add ~ 100 uL coated 0.5mm equilibrated glass beads to 1.5-2mL tube/well
  • Add 200 uL of yeast / fungus samples in UVT (or PCRopsis compatible buffer) to the tube with equilibrated beads
  • Vortex for ~5 minutes to produce lysate
  • Mix 20 uL lysate with 20 uL Reagent RVD – Enhancer (1:1 ratio)
  • Heat at 95C for 15 minutes then let cool for 10-20 seconds.

Extraction Time: ~15 mins

Fungal/Yeast-Dry nail clippings

  • Beat / crush dry nails in 200 uL of nuclease free water (or PCRopsis compatible buffer)
  • Add ~ 100 uL coated 0.5mm equilibrated glass beads to 1.5-2mL tube/well
  • Vortex for ~5 minutes to produce lysate
  • Mix 20 uL lysate with 20 uL Reagent RVD – Enhancer (1:1 ratio)
  • Heat at 95C for 15 minutes then let cool for 10-20 seconds.
